On April 27, 2020, Marsha Lyle had her last phone contact with her sister Bernadette, discussing her nephew's first steps. On May 3, 2020, authorities performed a welfare check at her home in Seale, Alabama, after her family grew concerned when she stopped answering calls and her mailbox became full. She appeared fine and was in bed. On May 5, 2020, a property manager found her in bed again but noted she was "not her normal self." On May 8, 2020, Marsha drove from Seale, Alabama toward Clewiston, Florida to visit her mother. At 12:56 p.m., she purchased gas at the Pilot Gas Station in Jasper, Florida. This was her last known debit card purchase. At 3:35 p.m., she was captured on security video entering a Walgreens pharmacy on Park Street in Jacksonville, Florida. At 6:28 p.m., she was seen on security video at a uniform shop on Phillips Highway in Jacksonville, walking away from her car. She never returned to the vehicle. Security footage showed her appearing to have aged drastically and not wearing her usual head covering. Two weeks later, on May 21, 2020, her vehicle, a white 2013 Toyota Yaris with Florida license plate 100QCV, was found abandoned in the parking lot of a shopping plaza on Phillips Highway near the uniform shop where she was last seen. She has never used her debit card or been heard from again.

On May 9, 2020, Marsha's mother filed a missing persons report with the Russell County Sheriff's Office in Alabama when she could not contact her daughter. The Jacksonville Sheriff's Office in Florida and the Russell County Sheriff's Office in Alabama have been investigating the case. On June 4, 2020, the Jacksonville Sheriff's Office announced that Marsha may be in the Jacksonville area. On July 9, 2020, police released a photo showing her in a Riverside pharmacy shortly after her disappearance, noting the differing hairstyles in different images. As of July 2020, the Jacksonville Sheriff's Office reported no updates in the case. By 2021, the family stated that the Jacksonville Sheriff's Office had made the case cold, citing insufficient leads. The family has conducted ongoing search efforts, driving from South Florida to Jacksonville every weekend to check homeless shelters and hospitals. Marsha had been diagnosed with Schizoaffective Disorder, which her family believed was caused by an inoperable brain tumor she had lived with for years. She had moved from Clewiston, Florida to Seale, Alabama in December 2019 to be closer to her church family. She was a devout Seventh Day Adventist and youth leader for the Pathfinders program at her church. She worked full-time at Marshalls and had taken two weeks off in September 2019 to help her sister with a newborn baby. Her family described her as modest, pure, kind, single, and devoted to her faith. She had a set schedule to call her sister Bernadette (who lives in California) and her mother Eileen Thompson (who lives in Clewiston, Florida) every Tuesday and Thursday.
